dc.description.abstractThis study investigated the effects ofextract of Launaeataraxacifolia (LT) on sub acute lead poisoning in adult rats, it evaluated the effects of one of dimerarptosuccinic acid (DMSA),Vitamin(VC); and combination therapy (DMSA + VC, DMSA + LT)on sub acute lead poisoning in adultrats. This was with a view to provide scientific basisfor the use of L. taraxacifoliain management of lead poisoning. The leaf of Launaeataraxacifoliawas extracted with methanol via cold extraction process.LD50 was determined, using Lorke method (Lorke, 1983). Sixty (60) rats of both sexes were randomly grouped into ten.One group was allowed free access to distilled water only, eight groups were allowed free access to lead acetate (2 mg/ml) in drinking water, for 5 weeks consecutively, seven groups out of the eight groups were later administered Launaeataraxacifolia(LT)extract (50, 100 and 200 mg/kg, p. o), DMSA, DMSA + LT, DMSA + VC and VC only for 21 days consecutively. The remaining group was administered LT extract (100 mg/kg, p. o.) for 21 days, and later allowed free access to lead acetate in drinking water (2mg/ml) for 5 weeks. Daily water intake, food intake, bodyweight and 24 - h urinary output (24-UO)were measured. Blood sample was collected and analysed on day 20 and 22 depending on the treatment. The following parameters were analysed: blood lead level (BLL), urine lead level (ULL), Alanine transaminase (ALT), Aspartate transaminase (AST), serum urea level (SUL), serumcreatinine level (SCL), serum albumin (SA), blood glucose, urine urea level (UUL), urine creatinine level (UCL), creatinine clearance (CC) and antioxidant activity [reduced gluthatione (GSH), catalase (CAT) and superoxide dismutase (SOD)]. Animals were further sacrificed; liver and kidney were harvested and processed for histological study. The oralLD50 was > 5000 mg/kg. The levels of (p < 0.05) BLL, ALT, AST, SUL, SCL and glucose were significantly decreased (p < 0.05) in the groups treated with LT. On the other hand, the levels of ULL, 24-UO, UUL, UCL and CCwere significantly decreased (p < 0.05) in groups treated with LT (p < 0.05). However, combination therapy (DMSA + VC and DMSA + LT) revealed marked improvement in BLL and CC than monotherapy (0.005 ± 0.002, 0.006 ± 0.002) and (7.3 ± 1.25, 7.3 ± 0.67) respectively. There was no change in food and water intake, bodyweight and SA. Meanwhile, GSH, CAT and SOD were significantly (p < 0.05) increased in groups treated with LT. Interestingly, the micrograph of liver and kidney revealed remarkable improvement in groups treated with LT. This study concluded that leaf of Launaeataraxacifolia has preventive and ameliorative effects on sub-acute lead poisoning. The extract also showed additive effect when combined with DMSA.en_US
